چکیده

Stefan Schulte-Merker has been working with zebra®sh for over ten years and was involved in a number of genetic screens. He was also part of a team that produced a ®rst-generation map of the `Goodfellow panel' of radiation hybrids, which was intended to help in the cloning of the mutated genes. He is currently Head of Fish Genetics at Artemis Pharmaceuticals, in TuÈbingen, who are in the process of a second-round genetic screen, called `TuÈbingen 2000'. Zebrafish: A Renewed Model System For Functional Genomics

In the post genome era, a major goal in molecular biology is to determine the function of the many thousands of genes present in the vertebrate genome. The zebrafish (Danio rerio) provides an almost ideal genetic model to identify the biological roles of these novel genes, in part because their embryos are transparent and develop rapidly. The zebrafish has many advantages over mouse for genome-...

Reverse genetics in zebrafish by TILLING.

TILLING, for Targeting Induced Local Lesions in Genomes, is a reverse genetics strategy that identifies mutations in specific genes of interest in chemically mutagenized populations. First described in 2000 for mutation detection in Arabidopsis, TILLING is now used in a wide range of plants including soybean, rice, barley and maize as well as for animal model systems, including Arabidopsis, Dro...

New ways to admire zebrafish: progress in functional genomics research methodology.

The main challenge of the post-genomic era is to functionally characterize genes identified by the genome sequencing projects. Model organisms, including zebrafish, are indispensable for this demanding task. Zebrafish has been successfully incorporated into large-scale genetic screens due to the optical clarity of the embryos and their accessibility to various experimental techniques throughout...

Morphants: A New Systematic Vertebrate Functional Genomics Approach

The vertebrate genome contains a predicted 50 000-100 000 genes, many of unknown function. The recent development of morpholino-based gene knock-down technology in zebrafish has opened the door to the genome-wide assignment of function based on sequence in a model vertebrate. This review describes technical aspects of morpholino use for functional genomics applications, including the potential ...

ZFIQ: a software package for zebrafish biology

UNLABELLED Rapid development, transparency and small size are the outstanding features of zebrafish that make it as an increasingly important vertebrate system for developmental biology, functional genomics, disease modeling and drug discovery. Zebrafish has been regarded as ideal animal specie for studying the relationship between genotype and phenotype, for pathway analysis and systems biolog...
